We're re-arranging the upstairs layout and the airing cupboard position is preventing us from our ideal layout. Is there any reason why the hot water cylinder shouldn't go up in the loft?
We're probably going for a pressurised system and obviously I would need to build a lagged enclosure around it but are there any other reasons why a hot water cylinder wouldn't be acceptable in the loft?

There's no argument against an UNVENTED cylinder in the loft - except the weight. You will need to create a firm base for it to stand on.

OTOH, jacking up a cold water storage tank to create a fall to a cylinder nearby is less likely to be worthwhile.

See also http://www.iphe.org.uk/databyte/tank_cistern_installation.pdf - a VERY cautionary tale! Be careful what you put over your head in case it ever falls on it.
