Expansion tank valve isn’t working

I need to top up boiler. I have done this many times by turning black valve whereby the water has quickly gone into system. However, there is nothing today. 2 weeks ago we had a power flush and I’m wondering if engineer has turned off a second valve which I’ve just noticed. It has a screw in centre but won’t budge.

1. That is not a usable valve, its a non-return valve. Don't undo it or you may have a bit of a flood!
2. Check that the handle on the usual valve (lower one to copper pipe in your photograph) is actually turning the spindle. The plastic handles have a habit of breaking, which gives the appearance of the valve opening and closing when in fact nothing is happening. Take the chrome screw out of the centre, remove the handle, and try turning the spindle with a small spanner, grips or pliers.

Actually, it is a usable valve, @Aigburth just don’t unscrew the bottom black screw. The top has a handle which should be turned with the lower one. It is a double check valve combined with a lever ball valve, very common valve used for filling loops.
