removal of fire place

I have removed a fire place but obviously need to block the hole left. is the best idea to secure plasterboard to a frame then skim the board? If so do I have to put a vent in the board as air will still come down the chimney?

Yes,you must have airflow circulation otherwise damp and musky smell at the bottom and the top cannot dried out from rain as there is no flow of air.
