I’m soon to be tiling our bathroom and it seems that anything less than 3mm is ill advised for non rectified tiles.

My father in law used 2mm spacers, but the grout line is closer to 4-5mm.

We want the thinnest we can have for cleaning reasons.

Also with a standard set adhesive, when do you remove the spacers? My father in law did it next day, but general advice seems to be before adhesive sets.

Quality grout or grout sealer won't cause you any problems. Our grout is tidy and nearly 20 years old. We do have house dehumidifiers and control moisture so no mould.
I pull packers before adhesive is set and clean off because some adhesives are waterproof and like concrete.
Other adhesives will clean off when dry.
