what membrane in garden room floor

Hi chaps, sorry it's another garden room one, but hoping one of you kind gents can help me please?

I've just finished my garden room base and fitted the joists (living the DIY dream).

I have to insulate it next, I plan to put 50mm foil back recticel first then 100mm rockwool rwa45? Should I put a vapour or breather membrane somewhere in the base? If so which and where? Osb3 is going on top of this when I do the floor.

Frame is 8x3 on ground screws and joists are 6x2.
