I currently have a Salus RT500. Two wires - Live (from the Potterton programmer in the airing cupboard) and a switched live to the Honeywell 2-port valve. It's 13 years old, works fine but I want to have the option to replace it if it bites the dust. Looking at the Netatmo thermostat as it gives the option for variable hysteresis and control through an app. The wiring diagram for the Netatmo says it uses two wires - COM and NO. Would I just wire the Live to COM and the switched live to NO?

Yes, as simple as that - the Netatmo and your original RT500 are both battery powered.
Although make sure the mains power to the boiler/wiring centre is still isolated before changing the connections over!

Alternatively, you have a world of smart thermostat options, by instead, changing your potterton programmer and decommissioning the RT500 and it's associated cable completely.
