If the valve is working and lets water out the top end of the flexi when the screw is turned then the check valve has to be stuck shut - the brass bit at the top that the flexi pipe connects to - or less likely, the check valve is on the wrong way, there should be an (spell edit) arrow on it, that should be pointing towards the copper pipe above it.
That's not a proper approved CH filling loop BTW rather something that's been cobbled together with normal plumbing parts and the check valve may have got gunged up. Can I suggest you get someone in to fit a proper filling loop, that will replace the screw valve at the bottom, the flexi and the double check valve at the top with a proper filling loop and that will also provide tap handles making it easier to use
