Surely the cable I've starred is the room stat as it's the only one going to the white of the 3 port valve! blue neutral yellow live to stat from programmer red switched live. You need to remove that cable and put a wire link between where the red wire and yellow wires came out. If you disconnect the cable you have marked you will be left with a live end!

The cable marked appears to be red to fire boiler yellow for pump controlled by boiler and blue neutral

Huzzah! I've not seen a migenie before, is it easy to use/ does it work well?

Hi Echo, yes its very easy to use. Its wireless and I can change the settings on the go which is very useful. There are others out there that are self-learning (hive and nest) but the MiGenie is more than whats required for me.
