I have a conventional system and was getting no hot water in the tank. The problem was the motorised valve which I moved manually and the water was then hot. The valve has now been replaced but the water in the tank is warm not hot. Also one of the manual isolation valves was replaced as it was leaking slightly.
Nothing has been touched on the boiler.
Any ideas on why the water is not getting hot enough?

Do you have an s plan y plan or other? You say the valve was renewed, was it the valve complete, valve body or actuator (powerhead)?

Immediate action drill...from army days
water warm not hot then heat cutting off at lower temp
what causes shut down? thermostat

No idea what red has to do with anything. Water is getting warm anyway, just not hot. Full valve replacement. No idea what s or y means. Water was hot with old valve manually put to hw, now only warm. Heating seems fine for the brief time I could stand it to be on.

Was the " manual isolation valve " a gate valve by any chance ?
If it was, could it have been a by pass ? Show us a pic showing its position on the pipework if need be.
Was the complete motorised valve changed including the brass body ?

Probably best posting a photo of said valve. You didn’t knock the cylinder thermostat down by any chance?

The complete motorised valve was replaced. At either side of this are two manual wheel type gate valves to isolate the motorised one. One of these was also replaced. Nothing else was touched. No changes to thermostats or anything on the boiler or anything else.

Couldn’t be that one or both gate valves haven’t opened fully, or have failed, eg snapped?

Is the motorised valve connected to 3 pipes or 2 ?
From what you have told us it sounds like you have a Y plan ,with mid position valve ( 3 pipes) is that so ?
